Pentosan polysulfate 3 mg/kg was given to knee osteoarthritis patients for 4 weeks. Scientists evaluated the efficacy of the medication before participant enrolment, every week for the first 4 weeks of treatment, and then at weeks 8, 12, 16, and 24.

The severity of joint stiffness, discomfort at rest, and pain when walking were among the seven assessments. A visual analog scale (VAS) was used to rate joint stiffness and discomfort while walking and resting. The 13 activities of daily living (ADLs) used to assess knee functionality include stair climbing and household duties.

 

Results and Conclusions

Pentosan polysulfate showed incredible efficacy. Compared to the placebo group, the continuation rates were more significant. The mean scores for the 13 ADLs were also higher in the pentosan polysulfate group, meaning they could carry out daily activities. This group also showed functional improvement from baseline at weeks 8 and 12.

The study concluded that 4 weekly injections of pentosan polysulfate were linked with a considerably shorter duration of joint stiffness and discomfort at rest compared to the placebo. In addition, in patients with knee osteoarthritis, this medicine also reduced pain when walking and overall function for 8 weeks after therapy ended.

Pentosan polysulfate may be beneficial in managing knee osteoarthritis symptoms, but it alone cannot treat the disease. Remember this medication is not yet FDA-approved for any conditions related to osteoarthritis in humans, and its benefits are under investigation.
